Introduction to structural bioinformatics; applications to blood coagulation factor V and factor VIII

Summary : Bioinformatics enables prediction of protein structures and functions and contributes to the design of new therapeutic compounds. These computer methods are also used to study DNA/RNA, polysaccharides, phospholipids or other small molecules. These theoretical approaches guide the design of experiments aiming to establish correlations between structure and activity; while assisting the analysis of experimental results. Blood coagulation involves formation of several macromolecular complexes. Proper functioning of this biological system depends on numerous protein/protein and protein/membrane interactions that are not well understood at present. We here describe some of the tools used in structural bioinformatics and show how to apply these concepts to coagulation factor V and factor VIII.
