Salicylates: what’s new in 2008? |

Summary : Salicylates remain to this day the most prescribed drug for inflammatory bowel disease (IBD) patients, despite the availability of new therapies such as immunosuppressant and anti-TNFs. 4- and 5-aminosalicylate acids (4-ASA and 5-ASA) are the active components of this ancient therapeutic class. In the last few years, considerable progress has been made in understanding salicylates mode of action. Anti-inflammatory and antitumour effects of 5-ASA are mediated by peroxysome proliferator-activated receptor gamma (PPAR-γ). These data support numerous retrospective studies suggesting that salicylates reduce the risk of colorectal cancer among patients with IBD. New galenic forms of 5-ASA, coated with a novel multi-matrix delivery system to achieve a sustained release of the drug throughout the colon, are being developed. These new forms have an easy dosing schedule and may improve patient compliance. |
