Methylation in haematological malignancies : techniques and prospects |

Summary : Epigenetics defines an heritable alteration in gene expression without any change in primary DNA sequence. Two fondamental mechanisms are involved in this regulation : the DNA methylation at cytosins bases within a CpG dinucleotide and histone modifications. The CpG island methylation located in the promoter region notably of tumor suppressor gene is associated with transcriptionnal inactivation and gene silencing. At the level of a cell, the transcritpome depends, in part, of methylation processes. The study methods of DNA methylation are based mainly on methylation study after chemical modifications of DNA by sodium bisulfite. Some of theses techniques are sensible, specific and applicable to the study of methylation in hematological malignancies. Aberrant DNA methylation may serve as a biomarkers for cancer detection, assessment of prognosis, prediction of response to therapy and a target for epigenetic therapy. |
