This review covers recent advances in Fas (CD 95) signaling. It describes the various phases leading to cell demise after Fas-triggering, and particularly focuses on formation of the Death inducing signaling complex (DISC), obligatory step before caspases activation. Relevance of alternative pathways are also discussed in view of a better understanding of pathologies related to Fas defect, as observed in patients with lymphoproliferative syndrome and auto-immunity.
