TLT-1 (TREM-Like Transcript-1) is a novel platelet membrane receptor which has been recently characterized and identified to play an important role in platelet biology. TLT-1 belongs to the Triggering Receptors Expressed on Myeloid cells (TREMs) family which includes several immunoreceptors expressed by innate immune cells. TLT-1 is exclusively expressed on megakaryocytes and platelets, sequestered in α-granules and rapidly brought to the surface of platelets upon activation. By linking fibrinogen and modulating calcium signaling, but also by displaying anti-inflammatory properties, TLT-1 appears to play an important role in maintaining vascular homeostasis, regulating both adhesion, aggregation and inflammation at sites of injury/infection. Recent evidences highlight that a better understanding of TLT-1 signaling and regulation, but also the capacity to modulate its activity, will certainly open new realistic therapeutic perspectives.
