Simultaneous high-performance liquid chromatography determination of four antiproteases in human plasma

Summary : A new HPLC assay was developed for the simultaneous determination of indinavir, nelfinavir, ritonavir, saquinavir, using 1 ml plasma sample. A liquid-liquid extraction procedure was coupled with a reverse-phase HPLC separation using column switching. Two phenyl columns (100 x 4.6 mm, and 250 x 4.6 mm) are commutated through a ten-port electrically actuated valve. The mobile phase contains acetonitrile and 0.03 M dinatrium hydrogenophosphate (55/45 %). The quantification is performed by UV-detection at a wavelength of 240 nm for ritonavir and saquinavir, and 261 nm for indinavir and nelfinavir. Calibration curves are linear over the range 0.5 to 20 mg/ml, 0.005 to 1 mg/ml, 0.05 to 10 mg/ml for ritonavir, saquinavir, indinavir and nelfinavir, respectively. Correlation coefficients are better than 0.998 for all compounds, with both within and day-to-day coefficients of variation lower than 10 %. This method is suitable for drug monitoring.
