Summary : Mechanisms of DNA repair play a crucial role in the preservation of genome integrity. They also allow to circumvent the damages induced by a large number of anticancer agents and thus contribute to clinical resistance to anticancer drugs. We describe here the origin of the main lesions occurring on the DNA molecule as well as the main mechanisms of DNA repair. We show in some examples how it may be possible to rationally inhibit the DNA repair pathways in order to improve the efficacy of the existing anticancer drugs. |
